How does Paroxetine work?

Paroxetine works by affecting the level of certain specific chemicals that are imbalanced in people with depression and other anxiety disorders.

Common side effects include asthenia, sweating, nausea, decreased appetite, drowsiness, dizziness, insomnia, tremors, nervousness, ejaculatory disturbance, and other male genital disorders.

• It can take 2-3 weeks for Paroxetine to start working.
• Do not stop treatment suddenly as this may cause an upset stomach, flu-like withdrawal symptoms, and sleep disturbance.
• If your doctor asks you to stop Paroxetine, you should reduce the dose slowly over 4 weeks.
• Avoid consuming alcohol when taking Paroxetine, as it may cause excessive drowsiness and calmness.
• The addiction/dependence potential of Paroxetine is very low.

Indications of Paroxetine

Paroxetine is used in the treatment of depression, Panic disorder, and anxiety disorder.

Pharmacology

The efficacy of Paroxetine, the active ingredient in Oxat, is presumed to be linked to potentiation of serotonergic activity in the central nervous system resulting from inhibition of neuronal reuptake of serotonin (5-hydroxy-tryptamine, 5 HT). Paroxetine blocks the uptake of serotonin into human platelets. It is a potent and highly selective inhibitor of neuronal serotonin reuptake and has only weak effects on norepinephrine and dopamine neuronal reuptake.

Dosage & Administration of Paroxetine

• Major Depressive Disorder: Initial dose of 20 mg/day, with possible increase up to 50 mg/day.
• Obsessive Compulsive Disorder: Initial dose of 20 mg/day, with possible increase up to 40 mg/day.
• Panic Disorder: Initial dose of 10 mg/day, with possible increase up to 60 mg/day.
• Social Anxiety Disorder: Initial dose of 20 mg/day.
• Generalized Anxiety Disorder: Initial dose of 20 mg/day.
• Posttraumatic Stress Disorder: Initial dose of 20 mg/day.

Dose adjustments should be made to maintain the patient on the lowest effective dosage, and patients should be periodically reassessed to determine the need for continued treatment.

Interaction

Interaction with tryptophan, drug-metabolizing enzyme inducers/inhibitors, alcohol, haloperidol, amylobarbitone, oxazepam, MAOIs, lithium, phenytoin, anticonvulsants, and warfarin.

Contraindications

• Concomitant use with MAOIs or thioridazine.
• Hypersensitivity to Paroxetine.

Pregnancy & Lactation

Use during pregnancy only if the potential benefit justifies the potential risk to the fetus. Caution is advised during nursing.

Precautions & Warnings

• Caution in patients with cardiac conditions, epilepsy, and seizures.
• Use with caution in elderly, debilitated patients, and those with severe renal or hepatic impairment.
